Quick summary: São Luís to Santo Amaro do Maranhão is ~225km of paved road and about 3h30 in an ordinary car — you don't need a 4×4 to get there. Shared vans do the run every day (~4h with the stops), with few departures, generally in the morning and early afternoon — anyone landing after ~2pm usually sleeps in São Luís or pays for a private transfer. At the entrance to the village there's a R$10 tourism fee per person, valid for 3 days, in cash. And bring plenty of cash: Santo Amaro has no ATM.
Anyone heading for Santo Amaro comes in from the other side of the park, without passing through Barreirinhas: it's the way to sleep inside the dune field with no boat and no connection.

About 225km in total (up to 235, depending on where you join the MA-320). The MA-320 has been paved and runs all the way to the village; an ordinary car gets in year-round. What changes is the state of the surface in the rain.
The shared van is the standard for anyone arriving by plane. Some companies pick up at Marechal Cunha Machado airport and at hotels — ask before you book. There are few departures a day and every company has its own timetable; a published time goes stale fast, which is why we don't pin one down here. Confirm the day before, or send us your flight and we'll tell you who's running on your day. With the stops, count on ~4h; the price varies by company and season — ask us.
A rental car works well here, unlike in Atins: the road reaches the village and an ordinary car does the job. It's worth it if you want freedom over your own schedule, or you're combining Santo Amaro with Barreirinhas. Inside the park, a good part of the tours are in accredited 4×4s belonging to the local cooperative — your car stays at the guesthouse.
A private transfer is worth it from 3 people up, or with big luggage — priced per car, fixed at booking.
At the entrance to the village, the tourism fee: R$10 per person, valid for 3 days, in cash. Exempt: residents, people with disabilities, under-12s and over-60s — check on the spot, the rule is municipal. There's no ATM; some guesthouses and the cooperative take Pix or card, but don't count on it.
The village sits inside the dune field: until June, the nearest lagoons are a 40min-1h30 walk away; from July on, as the water recedes, the 4×4 tours are the way to see the deep lagoons. It's all in the Santo Amaro guide.
You can combine them: it's ~96km of paved road between the two, 1h30 to 2h. The route runs back along the MA-320 to the junction and on along the BR-402 to Barreirinhas — the stretch with the most patching in 2026. There's no regular tourist service: there's a residents' van before dawn, Monday to Friday, that takes tourists; beyond that it's a private transfer, a chartered car or your own car, priced per car by arrangement. There are few vans back and most leave in the morning — confirm the departure the day before with whoever brought you. The rule: 3h30 on the road + 2h of margin. A van leaving at 7am puts you at the airport around 10:30am, with room to spare for afternoon flights. Morning flight? Sleep the night before in São Luís.
